PADDLE OVER DISK (USP 5)
Using the paddle-over-disk method, a transdermal patch is placed either between a glass disc and an inert PTFE mesh (APPFIVE-HR) or directly onto a stainless steel mesh
(APPFIVE-V35, APPFIVE-V56 and APPFIVE-V80, etc.). These are placed on the bottom of the vessel/glass jar, release side up, under the rotating paddle.
Transdermal tests are performed at 32ºC to simulate the temperature of the skin. Other variables such as elevation and sampling requirements are the same as for USP Method 2 (Paddle Method).
QLA's USP 5 products are compatible with all brands of dissolution testers.
Important:
The APPFIVE-HR watch glass patch holder is a borosilicate watch glass with a 17 mesh PTFE screen held together by "Kynar clips". Patches up to 90mm in diameter can be tested. It is compatible with all brands of dissolution testers and conforms exactly to the USP specification.
The patch holders APPFIVE-V35, APPFIVE-V56 and APPFIVE-V80 etc. have stainless steel mesh and can hold different patch sizes. Various mesh sizes are available. Mesh sizes not listed may be available on request.
